Human chorionic gonadotropin (hCG) is a glycoprotein hormone produced by the developing placenta shortly after fertilization. In normal pregnancy, hCG can be detected in both urine and serum as early as 7 to 10 days after conception (1-4). hCG levels continue to rise very rapidly, frequently exceeding 100 mIU/mL by the first missed menstrual period (2-4), and peaking in the 100,000-200,000 mIU/mL range about 10-12 weeks into pregnancy. The appearance of hCG in both urine and serum soon after conception, and its subsequent rapid rise in concentration during early gestational growth, make it an excellent marker for the early detection of pregnancy. The test utilizes a combination of monoclonal and polyclonal antibodies to selectively detect elevated levels of hCG in urine. At the level of claimed sensitivity, the hCG One Step Pregnancy Test Strip (Urine) shows no cross-reactivity interference from the structurally related glycoprotein hormones hFSH, hLH and hTSH at high physiological levels.

It increases just before a woman's most fertile day of the month. This LH increase triggers ovulation. During ovulation an egg is released from the ovary. Because the egg can be fertilized only 12 to 36 hours after ovulation, detecting ovulation in advance is very important.
You may do this test at any time of the day, but you should test at approximately the same time each day and reduce your liquid intake for 2 hours before testing.
To decide when to begin testing, determine the length of your normal menstrual cycle. The length of your cycle is from the beginning of one period to the beginning of the next (count the first day of bleeding or spotting as day 1). If your cycle length is irregular, that is, if it varies by more than a few days each month, take the average number of days for the last 3 months. The length of the menstrual cycle is the duration from your first menstrual bleeding day to the day before the next bleeding begins. Determine the length of menstrual cycle before you test. lf your cycle is shorter than 21 days or longer than 38 days, consult a physician. lf you do not know your cycle length, you may begin the test 11 days after your first period since the average cycle length is 28 days. Perform 1 test each day over a 5 days period, or until the LH surge has been detected.

How to Do the Test?
1.
Calculate the length of your normal menstrual cycle
Start counting on day one of your period (the first day of bleeding or spotting) — and stop counting on the first day of your next period. This is the length of your cycle. It could be anywhere from 20 to 45 days long.
NOTE:
If the length of your cycle is different each month by more than just a few days, then simply take the average number of days over the last three months.
If your cycle is irregular, you may want to use your shortest cycle length to determine when to test.
2. Determine the day to begin testing
Using the chart by finding the number that corresponds to the length of your cycle to help determine the day you should begin testing. If you do not know your cycle length, you may begin the test 11 days after your first period since the average cycle is 28 days.
Example: if you have a regular cycle of 28 days and start your period on the 4th day of the month, begin with the 4th as day one, count forward 12 days and begin testing on the 15th of the month.
3. When to test on a day?
It is fine to do the test at any time of the day but you should test at approximately the same time each day.
For best results, do the test twice a day, 10 hours or more apart, for a few days prior to the ovulation. First morning urine is not recommended as it is concentrated and may give a false positive result.
4. Before the Test
Reduce your liquid intake for two hours before testing as too much liquid will dilute the LH in your urine.
Read the instruction carefully before performing the test.
5. Do the Test
Step 1:
collect your urine in a clean, dry, plastic or glass container.
Step 2:
remove the testing kit from a foil pouch by tearing at the notch. Immerse the strip into the urine with arrow pointing towards the urine for 3 seconds.
IMPORTANT: do not allow the urine level to exceed the MAX (marker line), otherwise the test may not perform correctly.
Step 3:
take the strip out and lay it flat on a clean, dry, non-absorbent surface. Read the results within 5 minutes. Do not read results after 5 minutes.
Step 4:
throw the test card away once you’ve read the result.
Read the Results
1. Positive Result (LH Surge):
that means you will probably ovulate in the next 24-48 hours after the first positive ovulation test. It is the best time to have intercourse after 24 but before 48 hours if trying to get pregnant.
2. Negative Result (No LH Surge):
that means there is no LH surge and you should continue testing daily.
Reasons to get Negative Result

Start testing too early or too late in your cycle. Calculate your cycle and try not to miss your LH surge.
You have a short LH surge. Try testing twice per day for one cycle to avoid miss LH surge.
You didn’t ovulate. This can be due to a variety of factors like stress, illness or hormonal conditions. If you don’t get a positive result after a few months, or if your cycles are irregular, talk to your doctor.
The test has been performed incorrectly. Follow the instructions closely and use clean urine.
3. Invalid
It may be due to insufficient specimen volume or incorrect test performance. Repeat with a new test.
Factors that will affect the Result
* Women who are approaching menopause or taking fertility-enhancing drugs may not be able to rely on LH levels to predict ovulation.
* Some medicines including fertility treatments can affect the results of some tests.
* If you are recently pregnant, that may also affect the test results.
* A heavy intake of fluids prior to testing will dilute the hormone in your urine.
